Antaeus Labs' AXON is based on a triterpene known as Hederagenin -- extracted from Hedera Helix. This triterpene is a triple monoamine reuptake inhibitor: A high-affinity substrate for the dopamine, 5-HT, and norepinephrine neural membrane transporters. (DAT, SERT, and NET, respectively.) Before we expound on this specific function any further, a few general words on how these monoamine neurotransmitters work...
The human brain contains billions of neurons which release neurotransmitters. They all consist of a cell body (the soma) & a long chain (the axon) which connects via branches to other neurons. Transmission via the axon is how one cell can influence others, and this is often carried out via monoamines such as DA, NE, 5HT; in response to electrical stimuli, they are released from the axon terminal into the synaptic gap, where they seek out and bind to receptors on the other side of the gap, on other neurons. This binding is not permanent: most of these monoamines are shaken-off their receptors in a matter of milliseconds, and are then 'available' once again. Their function is terminated by monoamine reuptake from the synaptic cleft back into the neuron via membrane-bound transporter proteins. (Glycoproteins, typically.) Following reuptake, most are metabolized into inert derivatives, and some are recycled and re-released.
...As you can imagine, when reuptake proteins are inhibited, monoamine breakdown comes grinding to a halt and they start accumulating outside neurons -- eventually flooding synapses & increasing transmission to supraphysiological levels. That's the whole point. Reuptake-inhibition makes for a powerful stimulant. Although Hederagenin has its mechanism of action in common with some banned chemicals, no ingredient previously released as a supplement is anything quite like it. Ephedrine, for example, is a potent NET inhibitor, but has an extremely weak effect on dopaminergic transmission and no effects at all where the serotogenic pathway is concerned.
By increasing dopaminergic, adrenergic, and serotogenic transmission, you get an increase in energy levels, heightened focus, an elevated mood, increased confidence and drive, and an revved-up metabolic rate.

Where Pharmacokinetics Are Concerned:
Hederagenin is very lipophilic & nearly-insoluble in water. It is slowly and poorly absorbed from the GI tract, but the half-life & duration of effect of the absorbed fraction is long. (6-8+hrs.) As has been conclusively shown with another triterpene, boswelic acid, absorption and bioavailability will increase tremendously when taken with food.

Antaeus Labs: "Axon contains 60mg caffeine and 700mg Hedera Helix extract (standardized to 20% Hederagenin) per serving."
Caffeine is perhaps the single most ‘tried and tested’ sports supplement. The ergogenic effects of caffeine are extremely well documented, and include improved concentration and alertness, a reduction in the perceived effort during endurance exercise, increased endurance performance, and endurance performance capacity.
Hederagenin is considerably less famous, but it is an extremely interesting and potentially useful compound. For starters, oral administration of the triterpene Hederagenin has been demonstrated to have significant activity in rats and mice, which are believed to be a result of increased transmission of the monoamines serotonin, norepinephrine (noradrenaline) and dopamine. Indeed, extracellular levels of all of these hormones were significantly increased in the rat frontal cortex after oral administration of hederagenin at a dosage of 50mg/kg. The peak increases in 5-HT (serotonin), NE (norepinephrine), and DA (dopamine) were over 400% of baseline.

There's more: Because of the effectiveness of ivy-leaf extracts at treating coughs and catarrh, the β2 adrenergic effects of the monodesmoside α-Hederin (a water-soluble hederagenin glycoside also found in Hedera Helix extracts) have been studied.

Nutraplanet: "You've mentioned that hederagenin is a triterpene. I've heard that the bioavailability of triterpenes such as ursolic acid and oleanolic acid is very low. Has anything been done about this?"
Antaeus Labs: "Although the oral bioavailability of hederagenin is rightly described as ‘poor’ (probably between 0.8 and 2% when taken without dietary fats, and higher when taken with a meal) this is comparable to other similar triterpenoid compounds like ursolic acid. Hederagenin's bioavailability is low, but it's far from zero, and I believe that hederagenin's low bioavailability is offset by its extremely high affinity for monoamine transporters. In any case, this is the reason a normal serving of Axon contains over two grams of Hedera Helix extract.
Nutraplanet: "The Wikipedia page for hederagenin tells us that it's a pesticide. Is there any truth to this?"
Antaeus Labs: "That's true, and it probably puts some people off from the idea of taking it as a supplement – but it shouldn’t. Many other naturally-occurring plant-derived compounds have anti-microbial and anti-fungal properties. Resveratrol, for example, is one of the most popular supplements in both the sports and anti-aging circles, and has been successfully used as a topical pesticide. Caffeine is also a pesticide, as are many dozens of lesser-known herbal stimulants. These naturally-occurring compounds are safe for human consumption, and they should not be confused with synthetic organochloride insecticides. Hederagenin is not toxic to humans at any reasonable dose."

...Axon is really good. For some people it can give them the bad jitters.I can tell you right away its not CAFFEINE overload.So many claim that this thing is pure caffeine but this feels different. It actually gives you that motivation and helps with mood especially on days you are feeling real sore and too tired to do anything from yesterday's work out. This will keep you going. =)
